The RTL comes with the Garmin installed already, right? The OEM cable for the GPS connects to the sound system which drives the speakers.

Exactly what kind of device are you asking about ?
And what year and model of Spyder ? (I see that has been answered.)
The general answer is YES......but with '17 models and before it gets there via the GPS so 2-way phone won't work.....but other audio inputs will, if the Garmin is plugged in and turned ON. My phone will link up and I can hear incoming calls......but can't talk back.
For '18 and later......I have no idea because some of the answer depends on what device you are trying to link.
